Overview

Artemisia annua-specific T-cell receptors (TCRs) are specialized protein complexes on the surface of CD4+ T lymphocytes that recognize allergenic peptides from Artemisia annua (mugwort) pollen. These TCRs specifically interact with peptides, such as those from the major allergen Art v 1, when presented by Human Leukocyte Antigen (HLA) class II molecules like HLA-DRB1*01:01 [PMID: 31202634]. This interaction is the primary driver of the allergic immune response, triggering the activation and proliferation of Th2 cells which secrete cytokines like IL-4 and IL-13 [PMID: 28434685]. These cytokines subsequently promote IgE production and eosinophilic inflammation, leading to symptoms of allergic rhinitis and asthma. In therapeutic contexts, these TCRs are the functional targets of allergen-specific immunotherapy (AIT), which aims to induce immune tolerance and shift the response from a Th2 to a Th1 or regulatory T-cell profile [PMID: 30143314]. Understanding the specific TCR sequences and their binding affinities is crucial for developing targeted biologics and improving the efficacy of allergy treatments.
