Overview

Astrocyte elevated gene-1 (AEG-1), also known as metadherin (MTDH), is a multifunctional oncoprotein that plays a pivotal role in cancer progression, metastasis, and chemoresistance. Initially identified as an HIV-1-inducible gene in astrocytes, it is now recognized as being overexpressed in nearly all human cancers, where it correlates with poor clinical outcomes (Sarkar et al., 2009, Cancer Research). AEG-1 acts as a scaffold protein, integrating multiple signaling pathways including NF-κB, PI3K/Akt, and Wnt/β-catenin to promote cell survival, invasion, and angiogenesis (Emdad et al., 2013, Advances in Cancer Research). In the context of therapeutic development, AEG-1 mRNA is a primary target for gene-silencing technologies such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs), which aim to reduce the levels of the oncogenic protein (Fisher et al., 2012, Cell Cycle). Additionally, small molecule inhibitors are being explored to disrupt the interaction between AEG-1 and its functional partners, such as SND1, to inhibit its pro-tumorigenic activities (Hu et al., 2009, Cancer Cell). Targeting AEG-1 offers a potential strategy to sensitize tumors to chemotherapy and inhibit metastatic spread across a broad range of indications.
