Overview

ATP synthase F(0) complex subunit a is an integral membrane protein in the mitochondrial ATP synthase (Complex V) that forms part of the proton channel. Together with subunit c, it creates a pathway for proton translocation across the mitochondrial inner membrane. The resulting proton motive force enables the rotary catalytic mechanism of the F(1) portion of the enzyme, leading to the synthesis of ATP from ADP and inorganic phosphate. Subunit a is encoded by the mitochondrial genome (MT-ATP6) and is crucial for energy production in aerobic eukaryotes. Pathogenic mutations in the MT-ATP6 gene disrupt proton conductance, impairing cellular energy metabolism and causing mitochondrial diseases including neurodegeneration and cardiomyopathies[2][3][5][7][8].
