Overview

ATPase inhibitory factor 1 (ATPIF1), also known as ATP5IF1, is a nuclear-encoded mitochondrial protein that serves as an endogenous regulator of the F1F0-ATP synthase [1]. Its primary function is to inhibit the hydrolase activity of the ATP synthase when the mitochondrial membrane potential drops, such as during hypoxia or ischemia, thereby preventing the wasteful consumption of cellular ATP [2][3]. In many human cancers, ATPIF1 is significantly upregulated, which promotes the Warburg effect (aerobic glycolysis) and confers resistance to apoptosis by modulating mitochondrial structure and signaling [4][5]. Because of its role in tumor progression and chemoresistance, ATPIF1 mRNA is a target for therapeutic intervention using RNA interference (siRNA) or antisense oligonucleotides to restore normal mitochondrial function and sensitize cancer cells to treatment [6]. Additionally, the protein's role in protecting tissues during ischemic stress makes it a subject of interest for developing therapies for cardiovascular and neurodegenerative conditions [7].
