The B-cell-specific Moloney murine leukemia virus integration site 1 (BMI1) messenger RNA 3' untranslated region (UTR) is a critical regulatory segment of the BMI1 transcript that governs protein synthesis through post-transcriptional mechanisms. BMI1 itself is a core component of the Polycomb Repressive Complex 1 (PRC1), which is essential for the epigenetic silencing of tumor suppressor genes, most notably the CDKN2A locus encoding p16INK4a and p14ARF [PMID: 26437234]. The 3' UTR serves as a docking site for microRNAs and RNA-binding proteins that modulate mRNA stability and translation efficiency. In various malignancies, including acute myeloid leukemia and glioblastoma, BMI1 is frequently overexpressed, contributing to the maintenance of cancer stem cells and therapeutic resistance [PMID: 28235105]. Therapeutic strategies targeting this region, such as small molecules developed via the GEMS (Gene Expression Modulation by Small-molecules) platform, aim to selectively reduce BMI1 protein levels by interfering with its translation [PMID: 30104355]. This targeted downregulation is intended to exhaust the cancer stem cell pool and restore normal cell cycle control in aggressive tumors.
