Overview

Bacterial cell wall synthesis enzymes, specifically transglycosylases and transpeptidases, are essential for the construction and maintenance of the peptidoglycan layer—a critical component of most bacterial cell walls. These enzymes are central to bacterial growth, division, and survival. They catalyze the polymerization of glycan strands and cross-link peptide side chains, resulting in a covalently closed mesh-like structure called the peptidoglycan sacculus. These enzymes are primary targets for β-lactam antibiotics such as penicillin.

Mechanism of action

Inhibition of transpeptidase activity, leading to loss of cell wall integrity and osmotic lysis. Some drugs also inhibit transglycosylase activity or earlier steps in peptidoglycan synthesis.
