Overview

Bacterial pathogen-associated molecular patterns (PAMPs) and surface antigens from enteric bacteria represent a broad class of molecules essential for the survival and virulence of gut-dwelling pathogens such as Salmonella, Shigella, and pathogenic Escherichia coli. This heterogeneous group includes lipopolysaccharides (LPS), flagellin, porins, and capsular polysaccharides, which serve as the primary interface between the microbe and the host (Janeway et al., Immunobiology, 2001). Biologically, these molecules function as ligands for host Pattern Recognition Receptors (PRRs), such as Toll-like receptors (TLRs), which trigger the innate immune response and the production of pro-inflammatory cytokines (Mogensen, Clin. Microbiol. Rev., 2009). In disease states, the overstimulation of these pathways by enteric antigens can lead to severe conditions ranging from localized gastroenteritis to systemic septic shock (Raetz & Whitfield, Annu. Rev. Biochem., 2002). Therapeutic interventions include the use of polymyxins to sequester LPS, the development of polysaccharide-protein conjugate vaccines, and the use of monoclonal antibodies to neutralize specific surface components (Liu et al., Microbiol. Mol. Biol. Rev., 2014). Because this term describes a collection of diverse molecular entities rather than a single receptor or enzyme, it is often used as a categorical designation in immunological and clinical research.

Mechanism of action

Drugs and vaccines targeting these patterns work by neutralizing endotoxins to prevent inflammatory signaling, inducing active immunity through B-cell and T-cell activation, or providing passive immunity via opsonization and antibody-dependent phagocytosis (Di Giandomenico & Sellman, Curr. Opin. Microbiol., 2015).
