Overview

The Baculoviral IAP repeat-containing 5 (BIRC5) mRNA 3'-untranslated region (3'-UTR) is a pivotal regulatory sequence that controls the translation and stability of Survivin, a member of the inhibitor of apoptosis (IAP) family (UniProt: O15392). This region is specifically targeted by microRNA 483-3p (miR-483-3p), which typically functions to downregulate BIRC5 expression through post-transcriptional silencing (PMID: 25605117). In various malignancies, such as hepatocellular carcinoma and Wilms tumor, the disruption of this miR-483-3p/BIRC5 axis leads to Survivin overexpression, which facilitates tumor cell survival, inhibits apoptosis, and promotes chemoresistance (PMID: 21860413). Beyond BIRC5, miR-483-3p regulates other oncogenic or tumor-suppressive mRNAs, including IGF2 and PANDA, making the entire miR-483-3p-regulated transcriptome a complex therapeutic landscape. Therapeutic interventions targeting this axis include antisense oligonucleotides like LY2181308, which binds to BIRC5 mRNA to trigger its degradation, and experimental miR-483-3p mimics designed to restore natural growth-inhibitory pathways (PMID: 24510955).
