Overview

The Basic leucine zipper and W2 domain-containing protein 1 (BZW1) mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the BZW1 transcript that governs gene expression through post-transcriptional mechanisms. BZW1 itself is a protein involved in the regulation of translation initiation and is frequently overexpressed in various malignancies, where it promotes cell proliferation and epithelial-mesenchymal transition (EMT). The 3'-UTR contains multiple binding sites for microRNAs, such as miR-129-5p and miR-145, which normally act as tumor suppressors by limiting BZW1 production. In many cancers, the downregulation of these microRNAs leads to the stabilization and increased translation of BZW1 mRNA, driving tumor progression. Consequently, the BZW1 mRNA 3'-UTR is viewed as a significant therapeutic target for RNA-based interventions, including miRNA mimics and antisense oligonucleotides, aimed at restoring normal regulatory control and inhibiting oncogenic signaling. Research indicates that targeting this region can effectively reduce the metastatic potential and growth of cancer cells in preclinical models.

Mechanism of action

The 3'-UTR of BZW1 mRNA serves as a binding site for regulatory microRNAs (miRNAs) which induce translational repression or mRNA degradation. Therapeutic strategies involve using miRNA mimics to downregulate BZW1 expression or antisense oligonucleotides (ASOs) to interfere with regulatory protein binding, thereby inhibiting the oncogenic functions of BZW1 in tumor cells.
