BCL2 Associated X, Apoptosis Regulator (BAX)

Target
BAX
Molecular classification
Pro-apoptotic protein, Bcl-2 family member
01

Overview

BAX (BCL2 Associated X, Apoptosis Regulator) is a key pro-apoptotic member of the Bcl-2 gene family. It promotes programmed cell death by permeabilizing the mitochondrial outer membrane, leading to cytochrome c release and caspase activation. Its expression and activation are tightly regulated, and dysregulation of BAX function is implicated in cancer and other diseases. While BAX itself is not typically directly targeted by drugs, it is an important component of the apoptotic pathway and strategies exist to restore or enhance its pro-apoptotic function, particularly in cancer cells.

Mechanism of action

BAX itself is not directly targeted by drugs, but its activity can be modulated through targeting upstream regulators or downstream effectors in the apoptotic pathway. Some experimental drugs aim to restore BAX function in cancer cells.

Biological functions

ApoptosisRegulation of mitochondrial membrane permeabilityCell deathTumor suppression (indirectly, via p53-mediated apoptosis)

Disease associations

Cancer (colorectal cancer, T-cell ALL)Negative regulation of autophagy
05

Safety considerations

Excessive BAX activation can lead to unwanted apoptosis and tissue damage.
06

Biomarkers

BAX expression levels can be a biomarker for predicting response to chemotherapy or radiation therapy in some cancers.
