Os7BGlu26 is a specific beta-glucosidase enzyme found in Oryza sativa (rice) that belongs to the glycosyl hydrolase family 1 (GH1). This enzyme is primarily responsible for the hydrolysis of beta-D-glucosidic linkages in various substrates, including cellooligosaccharides and phenolic glucosides, which are critical for plant growth and development (Opassiri et al., 2006). By releasing active molecules from their glucose-conjugated storage forms, Os7BGlu26 plays a significant role in regulating phytohormone levels and activating defense-related compounds during biotic and abiotic stress (UniProt Q8H6H1). In the field of agricultural biotechnology, it is studied as a potential target for enhancing crop resilience and optimizing carbohydrate metabolism to improve yield. While it is not a target for human therapeutic intervention, its enzymatic activity is sensitive to common glycosidase inhibitors such as gluconolactone and 1-deoxynojirimycin, which are used to study its functional role in planta (Baiya et al., 2021). Understanding the structural basis of its substrate specificity allows for the design of targeted agricultural applications to modulate rice plant physiology.
The enzyme is targeted by competitive inhibitors that occupy the active site, preventing substrate binding, or by mechanism-based covalent inhibitors that react with the catalytic nucleophile to block the retaining double-displacement catalytic cycle (Opassiri et al., 2006).
