Overview

Beta-lactamases are enzymes produced by bacteria that confer resistance to beta-lactam antibiotics by hydrolyzing the beta-lactam ring. Ambler class B comprises the metallo-beta-lactamases (MBLs), which require one or two zinc ions at their active site for catalytic activity. MBLs can hydrolyze a broad range of beta-lactams, including penicillins, cephalosporins, and carbapenems, but not monobactams. They are not inhibited by classic beta-lactamase inhibitors, posing a significant therapeutic challenge.
