Class D beta-lactamases, also known as oxacillinases (OXA-type enzymes), are serine-based enzymes that hydrolyze beta-lactam antibiotics, conferring resistance. They are particularly problematic in pathogens like Acinetobacter baumannii and some Enterobacteriaceae, contributing to multidrug resistance. These enzymes inactivate beta-lactam antibiotics by hydrolyzing the beta-lactam ring, preventing the drugs from inhibiting penicillin-binding proteins (PBPs) essential for bacterial cell wall synthesis. Common variants include OXA-1/30, OXA-10, OXA-23, OXA-24/40, and OXA-48, which may exhibit different substrate specificities, including carbapenemase activity. Many clinically used beta-lactamase inhibitors are ineffective against Class D enzymes, making infections difficult to treat.
