Overview

Local vasoconstriction refers to the narrowing of blood vessels at a specific site, primarily due to the contraction of vascular smooth muscle cells (VSMCs) in the vessel wall. This process reduces the diameter of blood vessels, leading to decreased local blood flow and increased vascular resistance. It is a physiological mechanism involved in blood pressure regulation, blood flow distribution, thermoregulation, and hemostasis. The process is triggered by various stimuli, including neurotransmitters (e.g., norepinephrine), hormones (e.g., epinephrine, angiotensin II, vasopressin), and local factors. The primary effector cells are VSMCs, and contraction is mediated by increased intracellular calcium and activation of myosin light-chain kinase. Endothelial cells modulate VSMC tone through the release of vasodilators (e.g., nitric oxide) and vasoconstrictors (e.g., endothelin). While not a direct therapeutic target in itself, dysregulation of vasoconstriction contributes to diseases like hypertension and Raynaud's phenomenon, and it is harnessed therapeutically to reduce bleeding and prolong the effects of local anesthetics.
