Overview

BMI1 proto-oncogene, polycomb ring finger is a component of the Polycomb group (PcG) protein family serving as a key element of the Polycomb repressive complex 1 (PRC1)[6]. It mediates transcriptional repression by promoting chromatin compaction and maintaining the silenced state of developmental and cell cycle regulatory genes[4][6]. BMI1 acts as an oncogene by suppressing tumor suppressors such as p16 and p19, and is essential for stem cell self-renewal in multiple tissues, including hematopoietic and neural lineages[1][4]. BMI1 is implicated in several types of cancer, regulates genes involved in cell proliferation and DNA repair, and interacts with key signaling pathways (Wnt, Notch, Hedgehog, Akt, receptor tyrosine kinases)[1].
