BAMBI (BMP and activin membrane-bound inhibitor) is a transmembrane glycoprotein that serves as a pseudoreceptor for the TGF-beta superfamily, including TGF-beta, BMPs, and activins. It is structurally similar to TGF-beta type I receptors but lacks the intracellular kinase domain, allowing it to act as a dominant-negative inhibitor by sequestering ligands and preventing the formation of functional signaling complexes [1, 2]. BAMBI also plays a critical role in the Wnt/beta-catenin signaling pathway, often acting as a transcriptional target that promotes cell proliferation and the epithelial-mesenchymal transition (EMT) in various cancers [3]. In the context of fibrosis, BAMBI expression is typically downregulated, which removes the brake on TGF-beta signaling and leads to excessive extracellular matrix deposition [4]. Targeting BAMBI mRNA via RNA interference (siRNA) or antisense oligonucleotides is an emerging therapeutic strategy aimed at reducing its overexpression in malignancies like colorectal and gastric cancers to inhibit tumor growth and metastasis [3, 5]. Conversely, strategies to restore BAMBI expression or mimic its function are being explored for treating chronic fibrotic diseases of the liver and kidneys [4].
