Bone morphogenetic protein 2 (BMP-2) (BMP-2)

Bone morphogenetic protein 2 (BMP-2) is a potent osteoinductive cytokine belonging to the transforming growth factor-beta (TGF-β) superfamily (UniProt: P12643). It plays a fundamental role in bone and cartilage development by stimulating the differentiation of mesenchymal stem cells into osteoblasts through the activation of Smad-dependent and Smad-independent signaling pathways (PubMed: 26583358). In clinical medicine, recombinant human BMP-2 (rhBMP-2), such as Dibotermin alfa, is used as a therapeutic agent to facilitate bone repair in spinal fusions, non-union fractures, and maxillofacial surgeries (StatPearls: NBK557645). Despite its efficacy, BMP-2 therapy is associated with significant safety concerns, including heterotopic ossification, severe soft tissue edema, and potential immunogenicity (PubMed: 23778611). Furthermore, dysregulated BMP-2 signaling is implicated in various pathologies, including vascular calcification and certain malignancies, where it may influence tumor cell proliferation and metastasis.
