Overview

Bone morphogenetic protein receptor type-1A (BMPR1A) is a transmembrane serine/threonine kinase receptor that plays a critical role in the bone morphogenetic protein (BMP) signaling pathway, which is part of the larger transforming growth factor beta (TGF-β) superfamily. It functions as a cell surface receptor for several ligands, including BMP2, BMP4, GDF5, and GDF6, regulating osteoblast function, chondrocyte differentiation, and adipogenesis. Mutations or dysregulation in BMPR encoding genes are implicated in various diseases involving abnormal tissue development or homeostasis.

Mechanism of action

Ligand-induced assembly of heteromeric complexes between type I (such as BMPR1A) and type II BMP receptors at the plasma membrane. Type II kinases activate type I kinases through phosphorylation; activated BMPRIA then propagates signals primarily through phosphorylation of SMAD proteins that modulate gene transcription.
