Overview

The Bovine coronavirus spike protein (S protein) is the primary surface antigen of Bovine coronavirus (BCoV), a betacoronavirus that causes significant enteric and respiratory diseases in cattle [3, 13]. It is a large, trimeric class I fusion glycoprotein that forms the characteristic spikes on the viral envelope [3, 10]. The protein is composed of two subunits, S1 and S2, which are responsible for host cell attachment and membrane fusion, respectively [11, 12]. Specifically, the S1 subunit binds to 9-O-acetylated sialic acid receptors on the surface of respiratory and enteric epithelial cells [1, 14]. This binding event triggers conformational changes in the S2 subunit that lead to the fusion of the viral and host membranes, allowing the viral genome to enter the cell [3, 15]. As the major target for neutralizing antibodies, the spike protein is the central immunogen in commercial vaccines used to prevent neonatal calf diarrhea and winter dysentery [12, 14]. It also plays a critical role in the bovine respiratory disease complex (BRDC) by facilitating infection of the upper and lower respiratory tracts [11, 12]. Diagnostic assays, such as ELISA and immunofluorescence, frequently target the spike protein to detect BCoV infection in clinical samples [9, 13]. The protein's high degree of glycosylation and potential for antigenic drift present challenges for long-term vaccine efficacy and therapeutic development [11, 12]. Understanding the structural and functional properties of the BCoV spike protein is essential for developing next-generation antivirals and more effective immunization strategies [10, 15].
