The Breast cancer type 1 susceptibility protein (BRCA1) mRNA 3' untranslated region (UTR) is a critical regulatory segment of the BRCA1 transcript, spanning approximately 1.5 kilobases. It serves as a hub for post-transcriptional regulation, containing numerous binding sites for microRNAs (miRNAs) such as miR-146a, miR-182, and miR-17, as well as RNA-binding proteins like HuR. These interactions dictate the stability and translational efficiency of the BRCA1 mRNA, thereby controlling the levels of the BRCA1 protein, a vital tumor suppressor essential for DNA double-strand break repair via homologous recombination. Dysregulation of this region, often through miRNA overexpression or genetic variants (SNPs) that create or destroy miRNA binding sites, is a significant driver of "BRCAness" in sporadic and triple-negative breast cancers. Consequently, the BRCA1 3' UTR is an emerging therapeutic target for RNA-based interventions, including antisense oligonucleotides (ASOs) and miRNA inhibitors (antagomirs), aimed at restoring BRCA1 expression or sensitizing tumor cells to DNA-damaging agents like PARP inhibitors.
