Overview

Bromodomain testis-specific protein (BRDT) is a member of the Bromodomain and Extra-Terminal (BET) family of epigenetic readers, primarily expressed in the testes during spermatogenesis [1, 4]. It contains two tandem bromodomains, BD1 and BD2, which recognize and bind to acetylated lysine residues on histone tails, facilitating chromatin remodeling and transcriptional regulation essential for sperm development [4, 5]. BRDT BD2 specifically plays a critical role in the reorganization of chromatin during the transition from histones to protamines in spermiogenesis [2, 4]. Due to its restricted expression pattern, BRDT BD2 is a high-priority target for the development of non-hormonal male contraceptives [2]. Small molecule inhibitors targeting this domain aim to disrupt the interaction between BRDT and acetylated histones, leading to a reversible reduction in sperm production and motility [2]. Beyond contraception, BRDT has been implicated in certain cancers, such as NUT midline carcinoma and some leukemias, where its aberrant expression contributes to oncogenic transcriptional programs [3].

Competitive inhibition of the acetyl-lysine binding pocket of the second bromodomain, preventing the recruitment of the protein to acetylated histones and disrupting downstream transcriptional programs [2, 3].
