Overview

BACH1 is a transcription factor that functions primarily as a transcriptional repressor by forming heterodimers with small Maf proteins. It binds to Maf recognition elements (MAREs) in gene promoters to repress transcription of target genes such as heme oxygenase-1 (HO-1). BACH1 regulates oxidative stress response, heme homeostasis, cell cycle progression, hematopoiesis, and immunity. It is associated with breast cancer, Fanconi anemia, and metastasis. Recent studies highlight its role as a master regulator gene involved in redox biology, hypoxia response, and metastatic processes, making it an emerging therapeutic target for certain cancers.
