Overview

The hemolysin-coregulated proteins Hcp1, Hcp2, and Hcp6 are structural and effector proteins that form the central tube of the *Burkholderia pseudomallei* Type VI secretion system (T6SS). These proteins are secreted and can form hexameric structures critical for bacterial secretion of effectors involved in infection and bacterial competition. LolC is a membrane-associated component of the ATP-binding cassette (ABC) transporter system responsible for trafficking lipoproteins from the inner to the outer membrane in Gram-negative bacteria, necessary for cell envelope biogenesis and survival. All four antigens have been consistently identified as immunogenic in animal models and human studies, eliciting notable humoral and cellular immune responses after infection or vaccination. Recombinant protein vaccines using Hcp1, Hcp2, Hcp6, and LolC (alone or in combination) confer varying degrees of protection against lethal *B. pseudomallei* challenge in mice, supporting their role as promising vaccine candidates for melioidosis.
