Overview

The butyrophilin subfamily 3A (BTN3A) family consists of BTN3A1, BTN3A2, and BTN3A3, cell surface receptors belonging to the immunoglobulin superfamily and structurally related to the B7 family. BTN3A1 is best known for its intracellular B30.2 domain, which binds phosphoantigens and activates human Vγ9Vδ2 γδ T cells—a process involved in immune surveillance, infection control, and anti-tumor immunity. BTN3A2 and BTN3A3 function as co-regulators, enhancing BTN3A1 activity; BTN3A2 lacks the B30.2 domain. The BTN3A molecules are widely expressed in immune cells and epithelial tissues, with dynamic roles in T cell (both αβ and γδ) and NK cell regulation, as well as links to inflammation, infection, and cancer. Ongoing research targets these molecules for immunotherapy and cancer treatment[2][3][4][5][6][7][8].
