Dectin-1 (CLEC7A) and Dectin-2 (CLEC4N) are transmembrane pattern-recognition receptors (PRRs) belonging to the C-type lectin family, predominantly expressed on dendritic cells and macrophages (UniProt Q9BXN2, Q6EIG7). They serve as crucial sensors for fungal pathogens; Dectin-1 recognizes beta-1,3-glucans, while Dectin-2 recognizes high-mannose structures (PubMed 22972645). Activation of these receptors triggers the Syk-dependent signaling pathway, leading to the induction of Th1 and Th17 immune responses, which are essential for antifungal defense (Nature Reviews Immunology 2009). These pathways can also be leveraged in cancer vaccine strategies to enhance the immunogenicity of tumor antigens. In oncology, agonists like beta-glucans (e.g., Imprime PGG) are investigated for their ability to reprogram the immunosuppressive tumor microenvironment and enhance the efficacy of checkpoint inhibitors (Journal of Hematology & Oncology 2018). Beyond cancer and infection, dysregulated Dectin signaling is implicated in inflammatory and autoimmune conditions, such as psoriasis and rheumatoid arthritis. Therapeutic strategies targeting these receptors primarily focus on agonism to boost immunity, though antagonism may be relevant for chronic inflammatory diseases. However, because they are distinct proteins encoded by different genes, they are typically characterized as separate therapeutic targets despite their synergistic roles in innate immunity.
