Mechanism of action

PDE4A inhibitors work by binding to the catalytic site of the enzyme, preventing the hydrolysis of the second messenger cyclic adenosine monophosphate (cAMP) [Patsnap, NIH]. This leads to an accumulation of intracellular cAMP, which activates protein kinase A (PKA) and other downstream effectors [Patsnap, NIH]. The activation of these pathways results in the suppression of pro-inflammatory transcription factors like NF-kappaB and the reduction of inflammatory cytokines such as TNF-alpha, IL-17, and IL-23, while promoting the expression of anti-inflammatory mediators like IL-10 [Patsnap, NIH, Frontiers].
