Cancer-testis antigens (CTAs) are a family of proteins with highly restricted expression in normal adult tissues—primarily the testis—and aberrant re-expression in various cancers. NY-ESO-1, MAGE-A1, and MAGE-A3 are prominent members. NY-ESO-1 is one of the most studied and immunogenic CTAs, eliciting strong humoral and cellular immune responses, making it an attractive target for cancer immunotherapy through vaccine development and adoptive cell therapy. However, the heterogeneity of antigen expression within tumors can affect therapeutic efficacy.
