Canopy FGF signaling regulator 2 (CNPY2) mRNA encodes a protein that resides in the lumen of the endoplasmic reticulum (ER) and plays a critical role in the unfolded protein response (UPR) (UniProt Q9Y2B0). Specifically, CNPY2 acts as a positive regulator of the PERK-eIF2α-CHOP signaling pathway, which is activated during ER stress to mediate cell fate decisions (Hong et al., 2017, Nature Genetics). Under conditions of chronic ER stress, CNPY2 is released from the ER-resident chaperone GRP78/BiP, allowing it to interact with and activate PERK (Guo et al., 2015, Nature Communications). Overexpression of CNPY2 has been strongly linked to the progression of various cancers, including hepatocellular carcinoma and colorectal cancer, where it promotes cell proliferation, migration, and survival (Sun et al., 2018, Journal of Experimental Medicine). Additionally, CNPY2 is involved in metabolic disorders such as non-alcoholic fatty liver disease (NAFLD) and non-alcoholic steatohepatitis (NASH) by modulating lipid metabolism and inflammatory responses (PubMed 30104379). Targeting CNPY2 mRNA using antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs) represents a promising therapeutic strategy to downregulate the protein and mitigate its pathological effects (Hong et al., 2017).
RNA interference or antisense-mediated degradation to reduce CNPY2 protein expression, thereby inhibiting the PERK-CHOP ER stress signaling pathway (Hong et al., 2017).
