Capping actin protein, gelsolin like (CAPG) is a member of the gelsolin/villin family of actin-regulatory proteins encoded by the human CAPG gene. The protein controls actin filament architecture by reversibly capping the fast-growing (barbed) ends of actin filaments in a calcium and phosphoinositide-dependent manner, without severing the filaments. CAPG modulates actin-based motility, particularly in non-muscle cells such as macrophages, where it is enriched and important for processes involving membrane ruffling and cell movement. Its dynamic regulation enables rapid shifts in actin filament lengths in response to cellular signals. CAPG is structurally distinct from gelsolin in its lower calcium requirements and reversible capping function. Dysregulation and overexpression of CAPG are associated with aggressive tumor phenotypes and metastasis and may also be involved in inflammatory and immune processes
