Overview

Carbohydrate sulfotransferase 15 (CHST15), also known as GALNAC4S-6ST, is a Golgi-localized enzyme responsible for the biosynthesis of chondroitin sulfate E (CS-E), a highly sulfated glycosaminoglycan (UniProt Q9P2E5). CHST15 catalyzes the transfer of sulfate from 3-phosphoadenosine 5-phosphosulfate (PAPS) to the C-6 position of N-acetylgalactosamine 4-sulfate (PubMed: 11042166). CS-E plays a critical role in the extracellular matrix by modulating the activity of various growth factors and signaling molecules involved in inflammation, fibrosis, and tumor metastasis (PubMed: 26350150). Overexpression of CHST15 is associated with poor prognosis in several cancers, including pancreatic and esophageal cancer, as well as the progression of mucosal fibrosis in inflammatory bowel diseases like Crohn's disease (PubMed: 28343796). Therapeutic strategies targeting CHST15 mRNA, such as the siRNA drug STNM01, aim to silence the gene's expression to reduce CS-E accumulation and inhibit disease progression (ClinicalTrials.gov: NCT02908971). Clinical investigations have focused on the local administration of these RNA-based therapies to treat refractory intestinal fibrosis and advanced malignancies (PubMed: 30113345).
