CD66, also known as the Carcinoembryonic Antigen-related Cell Adhesion Molecule (CEACAM) family, consists of several highly glycosylated proteins (CD66a-e) belonging to the immunoglobulin superfamily (Source: UniProt). These molecules are predominantly expressed on the surface of myeloid cells, including granulocytes and their precursors, as well as various epithelial cells (Source: PubMed, PMID: 15141351). In hematologic malignancies, CD66 members—particularly CD66c (CEACAM6)—are frequently overexpressed on leukemic blasts in acute myeloid leukemia (AML) and acute lymphoblastic leukemia (ALL) (Source: Blood Journal). This expression pattern makes CD66 an attractive target for radioimmunotherapy (RIT), where monoclonal antibodies like besilesomab, labeled with radionuclides such as Yttrium-90, deliver targeted radiation to the bone marrow (Source: Journal of Nuclear Medicine). Such treatments are primarily employed as part of conditioning regimens prior to hematopoietic stem cell transplantation to eradicate residual leukemic cells while minimizing systemic toxicity (Source: StatPearls). Beyond oncology, CD66 is utilized in diagnostic imaging (immunoscintigraphy) to localize sites of infection and inflammation due to its high expression on activated neutrophils (Source: European Medicines Agency).
