Overview

A carrier protein in a conjugate is a highly immunogenic protein used to enhance the immune response against a linked antigen, typically a bacterial polysaccharide (Source: PubMed, PMID: 24035958). In conjugate vaccines, these proteins are essential because polysaccharides alone are T-cell independent antigens that do not elicit a robust immune response or long-lasting memory, especially in infants (Source: CDC, 2022). The carrier protein is internalized by B cells, processed, and its peptides are presented via MHC class II molecules to T-helper cells (Source: WHO, 2021). This interaction triggers B-cell differentiation into memory B cells and high-affinity antibody-producing plasma cells. Common examples of these proteins include CRM197 (a non-toxic mutant of diphtheria toxin), tetanus toxoid, and diphtheria toxoid (Source: StatPearls, 2023). While not a therapeutic target in the traditional sense of a receptor or enzyme, the carrier protein is a critical pharmacological component for the prevention of infectious diseases.

Mechanism of action

Carrier proteins provide T-cell epitopes that allow T-cell independent antigens to be recognized by T-helper cells, thereby inducing a T-cell dependent immune response characterized by isotype switching and immunological memory.
