Overview

The cartilage matrix, particularly in joint tissue (articular cartilage), is a specialized form of connective tissue that covers the ends of bones in synovial joints. Its primary function is to provide a smooth, lubricated surface for articulation and to facilitate the transmission of loads with minimal friction. The matrix is avascular, meaning it lacks blood vessels, nerves, and lymphatics. The ECM forms the bulk of cartilage and consists mainly of water (~65–80% by weight), collagen fibers (primarily type II collagen), proteoglycans (notably aggrecan), non-collagenous proteins, glycoproteins, and other minor components. It is organized into distinct zones (Superficial, Middle, Deep, Calcified) with functional regions around chondrocytes (Pericellular, Territorial, Interterritorial). Damage or degeneration of articular cartilage leads directly to conditions like osteoarthritis. Because it is avascular, self-repair capacity is limited. Recent advances include using bioactive peptides that mimic extracellular matrix motifs or bind specifically to components like type II collagen/chondrogenic ligands.
