Overview

Casein kinase I isoform gamma-3 (CSNK1G3) is a member of the casein kinase 1 (CK1) family of serine/threonine protein kinases, which are involved in a wide array of cellular processes including DNA repair, cell cycle progression, and signal transduction (UniProt: Q9Y6M4). CSNK1G3 specifically functions as a key regulator of the Wnt/beta-catenin signaling pathway, often acting to promote the stability and transcriptional activity of beta-catenin (PubMed: 21135508). It is also implicated in the regulation of the eukaryotic circadian clock by phosphorylating core clock proteins (PubMed: 15860454). In oncology, CSNK1G3 is frequently overexpressed in various malignancies, such as colorectal and breast cancers, where it contributes to tumor cell proliferation and survival (PubMed: 25605851). While several small-molecule inhibitors like D4476 and IC261 target the CK1 family, achieving isoform-specific inhibition for the gamma-3 variant remains a significant pharmacological challenge (PubChem). Therapeutic targeting of CSNK1G3 is being explored to disrupt oncogenic signaling, though potential side effects related to its role in normal tissue homeostasis and circadian regulation must be managed.
