The Caveolin-1 (CAV1) mRNA 3' untranslated region (3' UTR) is a critical regulatory segment of the CAV1 transcript that governs the stability and translational efficiency of the Caveolin-1 protein. Caveolin-1 is the principal structural component of caveolae, specialized plasma membrane invaginations involved in endocytosis, cholesterol homeostasis, and the regulation of various signaling pathways, including Src and Ras (UniProt P33176). The 3' UTR contains multiple binding sites for microRNAs (miRNAs) and RNA-binding proteins, such as miR-103, miR-107, and miR-204, which modulate CAV1 expression in response to cellular stress and environmental cues (Trajkovski et al., 2011; PubMed: 21654804). Dysregulation of the CAV1 3' UTR-mediated control is implicated in several pathologies; for instance, its downregulation is linked to pulmonary fibrosis and certain aggressive cancers, while its overexpression can promote metastasis in other contexts (PubMed: 22508264). Consequently, the CAV1 3' UTR is an emerging therapeutic target for RNA-based interventions, such as miRNA mimics or antisense oligonucleotides, aimed at restoring homeostatic Caveolin-1 levels in disease states.
