CCDC39 antisense RNA 1 (non-protein coding) (CCDC39-AS1)

Target
CCDC39-AS1
Molecular classification
Long non-coding RNA (lncRNA), Antisense RNA
01

Overview

CCDC39 antisense RNA 1 (CCDC39-AS1) is a long non-coding RNA gene located on chromosome 3 that functions as an antisense regulator of the CD39 (ENTPD1) gene by modulating its mRNA levels post-transcriptionally. It is enriched in regulatory T cells and Th17 cells from individuals with Crohn’s disease, and its inhibition using antisense oligonucleotides can restore CD39 expression and ameliorate disease activity in animal models. The molecule does not code for a protein and is not considered a traditional drug target, but represents a novel therapeutic target for RNA-based interventions in immune regulation disorders

02

Mechanism of action

For experimental oligonucleotide inhibitors, the mechanism of action is silencing of CCDC39-AS1, which upregulates CD39 expression, restoring immune regulation

03

Biological functions

Regulation of gene expression (post-transcriptional modulation of CD39/ENTPD1)Potential roles in neuronal identity and ciliary function (based on expression patterns)
04

Disease associations

Crohn’s disease (via regulation of CD39, which controls immune homeostasis)Currarino syndrome (gene-disease association, but mechanistic link unclear)Possibly other chronic inflammatory conditions linked to CD39 dysfunction
05

Safety considerations

No specific safety concerns are documented, but general concerns with RNA-targeting therapies (off-target effects, immunogenicity) would apply
06

Interacting drugs

FANA-CD39-AS
07

Biomarkers

Expression levels of CCDC39-AS1 in Treg and Th17 cells are associated with disease activity in Crohn's disease and potentially serve as disease biomarkers
