Overview

CD276 antigen, widely known as B7-H3, is a type I transmembrane glycoprotein and a member of the B7 family of immunomodulatory proteins (UniProt Q5ZPR3). While its mRNA is found in various tissues, protein expression is typically low in normal human cells but significantly upregulated in a broad range of solid tumors, including prostate, lung, and breast cancers (PMID: 31110138). B7-H3 primarily functions as a coinhibitory molecule that suppresses T-cell activation and cytokine production, thereby facilitating tumor immune evasion (PMID: 33537531). Beyond its role in immunology, it is involved in promoting tumor progression through pathways related to cell proliferation, migration, and angiogenesis. Due to its high differential expression between malignant and healthy tissues, B7-H3 has emerged as a prominent therapeutic target for monoclonal antibodies, antibody-drug conjugates (ADCs), and CAR-T cell therapies (PMID: 35121865). Current clinical efforts focus on leveraging B7-H3 to deliver cytotoxic payloads or to enhance the immune system's ability to recognize and destroy tumor cells.
