Overview

CD300 molecule-like family member F (CD300lf), also known as IREM-1, is a type I transmembrane glycoprotein primarily expressed on myeloid lineage cells such as monocytes, macrophages, and neutrophils (UniProt Q8TDQ1). It belongs to the CD300 family of immunoregulatory receptors and contains extracellular immunoglobulin-like domains and cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs) (Zenarruzabeitia et al., 2015). CD300lf functions as a rheostat for immune activation by recognizing ligands like phosphatidylserine and ceramide, which are exposed on apoptotic cells, thereby mediating efferocytosis and suppressing pro-inflammatory cytokine production (Simhadri et al., 2011). Beyond its role in homeostasis, CD300lf has gained significant attention as the essential cellular receptor for murine norovirus, where it dictates viral tropism and entry (Orchard et al., 2016). In drug development, CD300lf is considered a potential therapeutic target for treating inflammatory disorders and certain malignancies by modulating myeloid cell-mediated immunity. However, its broad expression across various immune cell subsets poses challenges for achieving cell-specific therapeutic effects without systemic toxicity.
