Overview

CD4+ T-cell receptors (TCRs) specific for Pediacel-derived peptide–MHC class II complexes are specialized immune receptors that recognize antigenic fragments from the Pediacel vaccine (Sanofi Pasteur, Pediacel Product Monograph, 2021). Pediacel is a combined vaccine protecting against diphtheria, tetanus, pertussis, polio, and Haemophilus influenzae type b (Hib). These TCRs are expressed on the surface of CD4+ T lymphocytes and bind to specific peptides derived from vaccine antigens, such as pertussis toxoid or tetanus toxoid, when they are presented by Major Histocompatibility Complex (MHC) class II molecules (Janeway's Immunobiology, 9th Edition). This interaction is the critical first step in the adaptive immune response, triggering T-cell activation, cytokine secretion, and the provision of help to B cells for high-affinity antibody production (PubMed: 21651135). Understanding the repertoire and frequency of these TCRs is vital for assessing the immunogenicity and long-term protective efficacy of the vaccine in pediatric populations. While these receptors are not targets for traditional inhibitory drugs, they are the functional targets of the vaccine itself, which acts as an agonist to stimulate protective immunity (PubMed: 11586998).
