Overview

CD4+ T helper cell activation via the PADRE epitope is a process involving the recognition of the synthetic PADRE peptide presented on MHC class II molecules of antigen-presenting cells (APCs) by the T cell receptor (TCR) of CD4+ T cells. This interaction, along with co-stimulatory signals, leads to T cell activation, proliferation, and differentiation into various effector subtypes, ultimately contributing to the adaptive immune response. PADRE is designed for broad HLA-DR binding, enhancing CD4+ T cell activation across diverse populations and serving as an immunological adjuvant in vaccine development. It enhances both cellular and humoral immunity against target antigens.

Mechanism of action

PADRE peptide binds to MHC Class II molecules on APCs, forming a complex recognized by the TCR on CD4+ T cells, leading to T cell activation, proliferation, and cytokine production. Co-stimulation via CD28/B7 is also required.
