Overview

The CD4 binding site is a highly conserved region on the HIV envelope glycoprotein gp120. This site is essential for the virus’s ability to attach to and enter host cells, specifically by binding to the CD4 receptor found primarily on helper T-cells. The interaction between gp120 and CD4 initiates a cascade of conformational changes in both gp120 and its partner subunit, gp41, ultimately leading to fusion of the viral membrane with the host cell membrane and entry of viral genetic material. Targeting this site is a major approach for developing neutralizing antibodies and small-molecule inhibitors to block viral entry.
