CD44 variant 6 (CD44v6) is a specific isoform of the CD44 transmembrane glycoprotein family produced by alternative splicing. While the standard CD44 isoform is widely distributed in various tissues, CD44v6 has a more restricted distribution. CD44v6 plays important roles in cellular adhesion, migration, and various signaling pathways. In cancer biology, CD44v6 has shown contradictory functions - in prostate cancer, its expression inversely correlates with disease progression and positively correlates with PSA-free survival, suggesting a less invasive phenotype. However, in other contexts, CD44v6 expression in non-metastatic rat carcinoma cells has been shown to convert them into metastatic cells and promote tumor progression. CD44v6 can also function as a co-receptor for the activation of growth-promoting tumor receptor tyrosine kinases. The standard CD44 and its variant isoforms like CD44v6 are involved in sensing and responding to changes in the tissue microenvironment[1][2][4].
