Overview

The CD45-derived peptide presented by HLA-A*03:01 is a specific peptide-major histocompatibility complex (pMHC) target utilized in the development of T-cell receptor (TCR) therapies for hematologic malignancies. CD45, also known as Protein Tyrosine Phosphatase Receptor Type C (PTPRC), is a transmembrane protein expressed on all nucleated cells of the hematopoietic system, making it a universal lineage-specific antigen. In this target configuration, a specific peptide fragment of the CD45 protein is presented on the cell surface by the HLA-A*03:01 allele, allowing for recognition by engineered T cells. This target is primarily utilized in the context of allogeneic hematopoietic stem cell transplantation (HCT), where TCR-T cells (such as TSC-102-A0301) are used to eliminate residual host leukemic cells and native hematopoietic cells. By selecting a donor who is HLA-A*03:01 negative, the therapy can selectively deplete the recipient's immune system while sparing the donor-derived graft. This approach aims to achieve complete donor chimerism and reduce the risk of relapse in patients with leukemia or myelodysplastic syndromes.
