Overview

Cytotoxic T lymphocyte (CTL) activation via the Class I Major Histocompatibility Complex (MHC-I) pathway is a central process in adaptive immunity, enabling the immune system to identify and eliminate cells infected by intracellular pathogens (such as viruses) or transformed into cancer cells. This pathway involves CD8+ T cells recognizing antigenic peptides presented on MHC-I molecules expressed by nearly all nucleated cells. Activation requires antigen recognition (TCR binding to peptide-MHC-I) and co-stimulation (e.g., CD28-CD80/86). Activated CTLs kill target cells through the release of cytolytic granules (perforin, granzymes) and Fas-FasL interaction.
