Photosan is a hematoporphyrin derivative (HpD) used as a photosensitizing agent in photodynamic therapy (PDT) (National Cancer Institute, 2023). Upon administration, the hydrophobic components of Photosan preferentially localize within cellular lipid bilayers, including the plasma membrane and the membranes of organelles like mitochondria and lysosomes (Journal of Photochemistry and Photobiology B, 2004). When these molecules are irradiated with light of a specific wavelength, they transition to an excited state and transfer energy to molecular oxygen, producing singlet oxygen and other reactive oxygen species (ROS) (CA: A Cancer Journal for Clinicians, 2011). Because singlet oxygen has a very short half-life and a limited diffusion radius of approximately 20 nm, the resulting oxidative damage is confined to the cellular macromolecules in the immediate vicinity of the membrane-localized Photosan (PubMed, PMID: 10335393). These targets primarily include unsaturated fatty acids in phospholipids, which undergo lipid peroxidation, and membrane-associated proteins, which may suffer from oxidation of amino acid residues or cross-linking (StatPearls, 2023). This localized destruction disrupts membrane integrity and triggers signaling pathways that lead to cell death, typically through apoptosis or necrosis, making it an effective strategy for treating various malignancies (PubMed, PMID: 15591000).
