Overview

The cellular membrane, composed of a phospholipid bilayer with embedded proteins and cholesterol, is a major site of oxidative damage. Reactive oxygen species (ROS) and reactive nitrogen species (RNS) can attack membrane lipids and proteins, leading to lipid peroxidation, protein oxidation, increased permeability, and ultimately, cell death. This process is implicated in a wide range of diseases and biological processes, including aging, cancer, and neurodegeneration. While cells possess antioxidant defense mechanisms, severe or prolonged oxidative stress can overwhelm these systems, leading to irreversible damage.
