Chemokine-like protein TAFA-4 (TAFA4), encoded by the FAM19A4 gene, is a secreted neurokine primarily expressed in the brain and C-fiber low-threshold mechanoreceptors (C-LTMRs) of the dorsal root ganglia [UniProt Q96LR4, Delfini et al., 2013]. It belongs to the TAFA family of highly conserved proteins that function as chemokine-like modulators of neuronal activity and sensory signaling [Tom Tang et al., 2004]. TAFA4 plays a pivotal role in the endogenous modulation of pain, specifically acting to inhibit mechanical and thermal hypersensitivity following tissue injury or peripheral nerve damage [Reynders et al., 2015]. Preclinical studies have demonstrated that the absence of TAFA4 leads to increased pain perception, while the exogenous administration of recombinant TAFA4 can effectively reverse mechanical allodynia in neuropathic and inflammatory pain models [Delfini et al., 2013, Kamitani et al., 2011]. Beyond its role in somatosensation, the FAM19A4 gene is a clinically significant biomarker in oncology, where its promoter methylation is used for the early detection of cervical and other epithelial cancers [De Strooper et al., 2014]. The TAFA4-related pathway represents a novel therapeutic axis for chronic pain management, aiming to restore sensory homeostasis without the side effects associated with traditional analgesics [Artic Therapeutics].
