Chitinase-3-like protein 1 (CHI3L1) mRNA encodes a secreted glycoprotein, also known as YKL-40, which belongs to the glycosyl hydrolase 18 family but lacks enzymatic activity (UniProt: P36222). This mRNA is highly expressed in various cell types, including macrophages, neutrophils, and cancer cells, particularly under inflammatory conditions (PubMed: 24501218). CHI3L1 plays a critical role in promoting cell proliferation, survival, and tissue remodeling while suppressing apoptosis and regulating the Th2 inflammatory response (PubMed: 30143548). In the context of disease, elevated levels of CHI3L1 mRNA and its protein product are associated with poor prognosis in several cancers, chronic obstructive pulmonary disease (COPD), and fibrotic disorders (NCBI Gene: 1116). Therapeutic strategies targeting CHI3L1 mRNA, such as antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs), aim to reduce the production of the YKL-40 protein to mitigate pathological inflammation and tumor progression (PubMed: 29760380). These RNA-targeted approaches are currently being explored in preclinical and early clinical research to address unmet needs in oncology and inflammatory diseases.
