Overview

The Choline kinase alpha (CHKA) mRNA 3'-untranslated region (3'-UTR) is a critical regulatory segment of the messenger RNA encoding the CHKA enzyme, which catalyzes the first step of the Kennedy pathway for phosphatidylcholine biosynthesis. This region contains multiple binding sites for microRNAs and RNA-binding proteins that dictate the stability and translational efficiency of the CHKA transcript (Source: PubMed, PMID: 26109445). In many human malignancies, including breast and lung cancers, CHKA is significantly overexpressed, contributing to increased cell proliferation, survival, and metabolic reprogramming (Source: NIH, Gene ID: 1119). Targeting the 3'-UTR specifically allows for the modulation of CHKA expression at the post-transcriptional level, offering a therapeutic strategy to deplete the enzyme and its oncogenic metabolic products. Experimental approaches include the use of antisense oligonucleotides and microRNA mimics designed to bind this region and trigger mRNA decay (Source: PMC, PMC4526088). By reducing CHKA levels, these interventions aim to disrupt tumor phospholipid metabolism and inhibit cancer progression.

Mechanism of action

Targeting the 3'-UTR of CHKA mRNA typically involves the use of antisense oligonucleotides (ASOs) or microRNA mimics to induce mRNA degradation or inhibit translation, thereby reducing the levels of the Choline kinase alpha protein and decreasing phosphocholine production.
