Overview

Chondroitin sulfate proteoglycan 4 (CSPG4), also widely known as NG2 or the pericyte-specific antigen, is a large type I transmembrane protein essential for microvascular stability and maturation (UniProt). It is primarily expressed on the surface of perivascular pericytes during active angiogenesis and is significantly overexpressed in several aggressive cancer types, including melanoma, glioblastoma, and triple-negative breast cancer (PubMed, PMC3969145). Biologically, CSPG4 functions as a co-receptor that facilitates cell adhesion and migration by linking the extracellular matrix to the cytoskeleton, while also stimulating pro-survival signaling through the MAPK and PI3K/Akt pathways (NIH). In oncology, CSPG4 is a high-value therapeutic target because its expression is largely restricted to activated pericytes and tumor cells, with minimal presence in quiescent adult tissues (PubMed). Current therapeutic strategies under development include chimeric antigen receptor (CAR) T-cell therapies and monoclonal antibodies designed to directly eliminate malignant cells and disrupt the tumor-supporting vascular network (ClinicalTrials.gov, NCT03073330).
